They are most effective when used together, because OPKs can tell you when ovulation is coming (usually) and BBT can confirm it happened. Basically, your negative OPK could mean several things. 1) You haven't gotten your surge yet. 2) You have gotten your surge (probably yesterday) and are ovulating today, but for whatever reason didn't ever see the positive OPK. Every cycle I have been able to measure the LH surge, and also the temp increase when I have ovulated and drop in BBT when AF is arriving. My FP/Ovulation has been all over the place though, so literally I measure OPK 2 times a day just to make sure I don't miss it.
